I'm sure Kool Moe Dee and Grand Master Caz are at least 2 MC's that flowed like that long before anybody from Long Island

I don't have any in hand that I could upload but if you locate any old Treacherous Three records you'll hear Kool Moe Dee rhyming that way. And if you can locate any old Cold Crush Brothers street tapes (those will be hard to find) you'll hear Caz at his finest (in my opinion). To me the Cold Crush brothers records released on Profile Records don't compare to their prior stuff.

It may depend what is meant by multi-syllable rhymes. There are several almost distinct styles of multi-syllable rhyming within rap and of course rappers did not invent this type of rhyming. Even before Rakim there was Lakim Shabazz and, yes, Kool Moe Dee.
A distinct style would be compounding words to make a rhyme just like the guy in ronnie2122 profile pic (Big L) did. I wouldn't know the official established rap artist to be the founder of Multi-Syllable rhymes. I think one might argue inventors of signature rhymes styles involving the use of multi-syllables.

Short answer impossible to know who was first.
I'll look for the one later.I asked this question before and I just find the more you search back in history you find others prior maybe not as established as the marquee names, but know for multi's.

Rakim,Kool G Rap, Kool Moe Dee would all be pioneers of the multi's.

I have no idea the so called First One. Even Kurtis blow in his 70's-80's music occasionally uses multi's although known for his monosyllables.

To be very brief I just wanted to point out the different styles of multi's.
i.e Really and silly (one word)
i.e [One style/Sundial] [All eyes/Multi's,Salt Fries] [Round House, Down South] (compounding words to make multis)
And there can be endless variations of rhymes schemes and syllable counts.
